Effects of Sulfaguanidine upon Hepatic Injury Produced by Carbon Tetrachloride in Rats

Abstract
The experiment was carried out to clarify the intriguing "protective" effects of sulfaguanidine on ultrastructural alterations in the hepatic cells exposed to carbon tetrachloride.
The carbon tetrachloride, sulfaguanidine, or both two drugs were administered into the albino rats and hepatic lesions were examined histopathologically and electron microscopically with following conclusions.
1. Hepatic tissue from animals receiving carbon tetrachloride revealed marked degenerative changes and frank necrosis in the central zone of lobules, but the hepatic tissue of animals which received both carbon tetrachloride and sulfaguanidine revealed no such alterations except three cases out of nine, which revealed minimal degenerative changes. The hepatic tissue from the animals fed only with basal diet or received sulfaguanidine alone revealed no significant pathologic changes.
2. Hepatic cells affected to carbon tetrachloride revealed wide spectrum of ultrastructural changes. The nucleus appeared active and the rough-surfaced endoplasmic reticulum revealed variable changes evidenced by swelling of these organelles and shedding of ribosomes. The mitochondria later underwent swelling with loss of cristae.
In contrast to animals receiving carton tetrachloride alone, animals received both carbon tetrachloride and sulfaguanidine revealed no such alterations in most cases but in some cases revealed only minilmal changes in endoplasmic reticulum such as mild dilatation and shedding of ribosomes.
3. Sulfaguanidine, therefore, was thought to have strong protective effects upon hepatic injury produced by carbon tetrachloride.
